  • ABSTRACT Nifedipine, a calcium channel blocking agent, is widely used in medical practice for the management of cardiovascular disorders recently. On the other hand, this drug has several side effects including gingival hyperplasia and increased risk of cancer. This study was conducted to examine the influence of nifedipine on gingival hyperplasia and the expression of p53 protein in those tissue. Forty-five male Sprague Dawley rats were randomly divided into 15 groups. Animals in each group daily received nifedipine in dimethyl suLfoxide at a dosage of 0 (control), 15, 30 and 60 mg/kg body weight for 1, 3 and 6 weeks. Animals were sacrificed at the end of the study. Gingival tissue blocks were processed immunohistochemically analized for the presence of p53 protein. Gingival gross morphology was assessed from stone models obtained from the mandibular incisal region. The results of the study showed that nifedipine induced gingival hyperplasia. Gingival dimensions including bucco-lingual and mesio-distal width and also apico-coronal height significantly increased following nifedipine administration either in dosage and treatment duration (p
