Production and characterization of monoclonal antibodies for detection of virulent factors of streptococcus suis

  • The muramidase released proteins and the extracellular factors of S. suis were isolated from bacterial surface by lysozyme treatments or precipitated from culture supernatant by ammonium sulphate and analyzed using sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot. Monoclonal antibodies against MRP and polyclonal antibody against EF were produced to differentiate the virulence of S. suis from avirulent strains using a reference strain D282. All bacterial cultures used in this study were identified as S. suis. Serotyping of the S. suis isolates revealed capsular types 1, 2, 4, and 11 and several cultures were non typeable. Among the 10 S. suis isolates used in this study, there were 2 isolates from infected pigs reacted with monoclonal antibody against MRP (136,000) and one reacted with polyclonal EF (110,000), representing phenotype MRP+EF+ and MRP+EF-. These phenotypes belong to S. suis serotype 2. Protein preparation of the remaining cultures did not react with both antibodies. This study indicated that MRP and EF could be identified in S. suis serotype 2. Keywords: monoclonal antibody -- MRP â�� EF â�� Streptococcus suis
