- The influence of ATP and vitamin E supplementation on the erythrocyte fragility test of CPD blood in vitro were studied. During this study, blood samples were collected from 10 blood donors in Indonesian Red Cross (PMI) Yogyakarta branch. Each samples were devided into 10 aliquots and each aliquot were devided into 4 tubes. Aliquot Ito VII were used as treatment samples, whereas aliquot VIII to X were used as control samples. An amount of 1 mg/ml blood ATP, 2 mg/ml blood, 4 mg/ml blood, was added to aliquot I, II and III respectively. An amount of 0,02 mg/ml blood vitamin E was added to aliquot IV. One mg ATP and 0,02 mg vitamin E per ml blood was added to aliquot V and 2 mg ATP ang 0,02 mg vitamin E per ml blood was added in aliquot VI In aliquot VII was added 4 mg ATP and 0,02 mg vitamin E per ml blood. Into the control samples, an appropriate volume of water, ethanol and a mixture of ethanol-water were added to aliquot WIZ, IX and X respectively. All the samples were then kept in refrigerator at 40*- 6°C for four weeks. Erythrocyte fragility test using Baker and Silverton method was carried out every week. The result shows that ATP supplementation decreases the fragility of erythrocyte. Similar result was observed in samples with vitamin E supplementation. When both ATP and vitamin E were used the higher the ATP concentration the lower the erythrocyte fragility. Comparing the erythrocyte. fragility in the supplementation of ATP and a mixture of ATP and vitamin E, the result shows that the latter decreases erythrocyte fragility significantly than that of the former. Samples with vitamin E supplementation shows no difference in the fraagility erythrocyte with a mixture ATP and vitamin E supplementation in the first week. However significant difference was observed in the secod, third and fourth week. This study shows that either ATP or vitamin E supplementation decreases erythrocyte fragility of CPD blood donors in vitro. In addition, more significant decrese in erythrocyte fragility is observed when both ATP and vitamin E were supplemented than those with ATP or vitamin E.
