- The isolation of plant resistance (R) genes provides opportunities to produce crop plant varieties with increased disease resistance. Previous techniques to clone plant R genes, including transposon tagging and map base cloning, are lime consuming and may require a knowledge of a location of R gene in the genome. A new technique called Representational Difference Analysis (RDA) offers a straightforward method for cloning DNA fragments differences between tivo complex genomes. In this study, the technique was used to clone a DNA fragment found only in tomato plants CLycopersicon esculentura) which are resistant to Altemaria stem canker. This suggests that RDA could provide a rapid method of cloning disease resistance genes in plants.
