Effect of glucose addition into capacitation medium for beef cattle spermatozoa on in vitro fertilization process

  • A research was conducted to study the effect of glucose addition into capacitation medium for beef cattle spermatozoa on in vitro fertilization process. Oocytes were collected from slaughter-house bovine ovaries. Frozen-thawed spermatozoa were used in the present study. Two kinds of media were used for capacitation, B-O with 5mM glucose and B-O without glucose. Sperm capacitation was evaluated indirectly using parameters of fertilization and cleavage rates. Data were analyzed statistically using X2~test. Fertilization rate revealed from the two media was 21 % and 24 %, while cleavage rate was 34.6 % and 34.5 %, respectively. Results of the present study showed no significant difference in both fertilization and cleavage rates between glucose and non-glucose added media. In general, 2 cell-stages were observed after 48 hours post fertilization and few of them cleaved after 24 hours post fertilization. It can be concluded that the addition of glucose in capacitation medium for beef cattle spermatozoa does not effect the fertilization and cleavage rates in the in vitro fertilization process.
