- ABSTRAK Pasteurella multocida serotype B:2 causes hemorrhagic septicaemia, an acute disease affecting cattle. Hemagglutinin was suspected to take part in pathogenesis process. Recombinant plasmid pKH400 had an insert of DNA fragment about 5 kb containing hemagglutinin 32 kDa gene of Pasteurella multocida. The purpose of this experiment was to determine the location and restriction map of hemagglutinin gene of pili subunit Pasteurella multocida B:2 at pKH400. The stages of experiment include restriction mapping of pKH400, followed by subcloning DNA fragment into an expression vector. The result of gene expression was analysed by hemagglutination test and western blotting using polyclonal antibodies against hemagglutinin. The result showed that hemagglutinin 32 kDa gene was located in the Hzndlll-HmdIII fragment (1,5 kb) of inserted DNA pKH400 (pKH405). The 1,5 kb Hindlll-Hindia DNA fragment had one restriction site of EcoRI. Hemagglutinin gene posses putative promoter which is recognized by RNA polimerase Escherichia coli. Subcloned-hemagglutinin 32 kDa protein was recognized by antibody against hemaglutinin 32 kDa. Keywords: hemagglutinin -- Pasteurella multocida � subcloning, restriction mapping
