- The objective of the study is to determine antibacterial and antibiofilm activity of ethanolic extracts and methanol solublefraction from the bulbs of tiwai (Eleutherine americana. Merr) against Staphylococcus aureusATCC 25923 and it's clinical isolates (SA 28, SA 913, SA 944, SA 947, SA 957 and SA 758). The contained compound in samples were also identified in this study. Antibacterial activity testing was conducted using macrodilution method while the inhibition of biofilm formation and it�s degradation were calculated using tissue culture plate method with 0.1% crystal violet as detection end point. MIC range from ethanolic extract and the methanol soluble fraction respectively of 0.5 - 1 mg/mL and 0.25 - 0.5 mg/mL while MBC can only be achieved by methanol soluble fraction at concentration 2 mg/mL on S.aureus ATCC 25923 and SA 758. Ethanolic extract at subinhibitor MIC can inhibit biofilm formation on tested bacteria except SA 913 and SA 944 while methanol soluble fraction except SA 913. Methanol soluble fraction at concentration 0.01 - 0.5 mg/mL can degrade biofilms from all tested bacteria while the ethanolic extract can only degrade the biofilms on SA 28, SA 947, SA 957 and ATCC 25923. Samples at subinhibitor MIC concentrations may also increase the formation of biofilms in the tested bacteria.Total phenolic concentration from ethanol extract and methanol soluble fraction respectively 52,74 ± 0,31%GAE and 70,31 ± 0,40%GAE. hRx value from 3 spot detected as naphthoquinone with lawson as reference standar respectively 100 (hRf 92), 78(hRf 75) and 46 (hRf 46). Both samples show antibacterial and antibiofilm activity on bacteria tested, especially on SA 758. Samples can also induce biofilm formation in a certain concentration.
