  • Methotrexate is a key drug in acute lymphoblastic leukemia (ALL) treatment. It inhibits DNA replication by blocking the conversion of 5,10 methylenetetrahydrofolate to 5-methylenetetrahydrofolate by methylene tetrahydrofolate reductase, a critical enzyme involved in folate metabolism, DNA methylation and synthesis. It has two common functional polymorphism (C677>T and A1298>C). Genetic polymorphisms of this enzyme have been shown to impact relapse risk in childhood ALL. This study aimed to asses the impact of MTHFR polymorphisms C677>T on the relapse of childhood ALL. This case-control study was undertaken to analyze the presence of the C677>T mutant alleles in 36 (relapse group) and 36 (no relapse group) childhood ALL patients. Polymorphisms in MTHFR C677>T were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).
