  • Periodontal disease is an inflammation, degeneration of soft tissue and the teeth bone supporting that have high prevalence and intensity. One of periodontal disease is periodontitis. Periodontitis causes bone destruction by the interaction between the bacteria and the host that causes bone resorption by osteoclasts. Alveolar bone destruction which caused by periodontitis can be treated with topical application of simvastatin. Simvastatin is a statin drug that has antiinflammatory, immunomodulatory, antioxidant, antithrombotic, and osteoblastic differentiation. The purpose of this research is to determine the effect of simvastatin topically to the amount of osteoclast ratâs bone with periodontitis. Twenty four Sprague dawley rats were divided into a treatment group (simvastatin gel) and the control group (CMC-Na gel). All rats induced periodontitis using steel ligature diameter of 0.4 mm were associated around the mandibular incisors. Seven days after the induction of periodontitis, all rats given the treatment gel application on day 0 to day 14 every day. In calculating, the reduction in osteoclast appear from day 3 to day 14. The result of decrease in the average number of osteoclasts larger in treatment group than the control group as shown in the 5th to the 7th day. 4.22 in the treatment group on day 5 and 1.88 on day 7, whereas the control group 6,56 on day 5 to 6.11 on day 7. Two way ANAVA showed that p = 0.00 (p> 0.05), can be interpreted that occur decline in the number of osteoclasts significantly. From this research, it was concluded that the topical application of simvastatin can decrease the number of osteoclasts in periodontitis.
