- Indigenous yeasts of Pichia manshurica and Rhodosporidium paludigenum were able to produce inulinase enzyme, but the activity relatively low. The aims of the current researches were to improve strain quality inulinase production through enviromenthal conditions and genetics manipulations. One of the techniques used to improvement yeast strain was protoplast fusion technique. Protoplast fusion process could be done through using intraspecific, interspecific and intergeneric involving two or more parental genomes. Protoplast fusion can be performed through several stages namely: isolation of protoplast, protoplast fusion, regeneration of fusion protoplast and analysis of hybrids (fusants). Dahlia (Dahliasp.) tubers were used as an inulin source in the research. The inulin stimulated yeast to produce inulinase enzyme (EC. 3.2.1.7).
