  • Ketoprofen, commonly used in rheumatoid arthritis treatment, has low solubility and cause gastritis while using orally. Solid SNEDDS can overcome this problem by forming an o/w nanoemulsion in gastric fluid. The purpose of this study was to make a solid SNEDDS using oleic acid, tween 20, propylene glycol, and aerosil. Optimization was done by simplex lattice design. The clarity of nanoemulsions, the emulsification time, and the AGF/AIF stability of SNEDSS optimum formula were carried out. The optimum formula incorporated with aerosil to obtain a solid SNEDDS by spray drying and solid carrier adsorption method. The clarity of nanoemulsion, drug content, and powder morphology of solid SNEDDS were evaluated in this study. The optimum formula of SNEDDS ketoprofen consists of 0.714 mL oleic acid, 4.107 mL tween 20, and 0.179 mL propylene glycol. The particle size analysis results showed that the average diameter of nanoemulsion droplets size is 25.6ï�±7.4 nm, with 99.67ï�±0.321 %T clarity and 36.04ï�±1.25 seconds emulsification time. The SNEDDS formula is stable in AGF and AIF. The solid SNEDDS powder formed nanoemulsion with clarity 77.03ï�±5.82 %T (spray drying method) and 99.4ï�±0.4 %T (solid carrier adsorption method), drug content 53.94ï�±2.57 % (spray drying method) and 53.41ï�±2.52 % (solid carrier adsorption method). The solid SNEDDS powder from spray drying method has a spheris morphology, while the powder from solid carrier adsorption method has an amorf morphology.
