- Milking process seems to be stressful for dairy cows seen from an increase in cortisol. Normally, milking machine could release oxytocin and milk ejection reflex via neuroendocrine, but sometimes could inflict stress and causing substraction of milk production. Hand milking is still widely use in Indonesia, even though there are several dairy farms already used milking machine. The purpose of this study was to determine the physiological and behavioral responses of dairy cows due to milking parlor portable. Research used 20 multiparous lactating dairy cows aged 3-6 years from Balai Besar Pembibitan Ternak UnggulHijauan Pakan Ternak (BBPTU-HPT) Baturraden, Purwokerto, Central Java. Blood sampling were collected 3 times, before milking machines were installed, 50 minutes and 100 minutes after milking process started. Assay cortisol used Enzyme Linked Immunosorbent Assay (ELISA) method. Comparison of the ratio N/L is performed with differential leukocytes. Statistical analysis used one-way ANOVA. Blood serum cortisol concentrations results are as follows: before milking: 7,27 ± 4,64 ng/ml
