- Synthesis and antibacterial In-Vitro test of N-phenyl pirazoline compound from benzldehyde and vanillin with acetophenone have been carried out. The research has been conducted via several steps of reactionsm i.e.: protection of hydroxyl group on vanillin with benzoyl cloride, chalcone synthesis from benzaldehyde and vanillin with acetophenone and pirazoline synthesis of the related chalcone with phenylhidrazine. Finally, chalcones and pirazolines were tested as an antibacterial agent by In-vtro test. Protection of hydroxyl group on vanilin was carried out by esterification with benzoil cloride using sonochemistry methode. Synthesis of benzaldehyde chalcone was performed by stirring benzaldehyde with acetophenone for 2 hours, while vanillin chalcone was synthesied by heating vanillin and acetophenone with methanol at 60-70 oC. Pirazolines synthesis was done by refluxing the related cahlcone with phenylhydrazine in acidic condition for 4 hours. Structure elucidation of all products was confirmed by FT-IR, 1H-NMR, 13C-NMR and GCMS spectrometers. Antibacterial test of chalcones and pirazolines was conducted by cup-plate technique methode againts Staphylococcus aureus, Bacillus subtillis, Bacillus cereus, Escherechia coli and Shigella flexneri. The result showed that benzoyl vanillin was obtained as white solid in 92.45% yields by sonochemistry methode 5 minutes reaction. Aldol condensation produced benzaldehyde chalcone as white yellowish solid in 47.04% and vanillin chalcone as yellow solid in 52.63% yields. Cyclo-condensation of the related chalcone with phenylhydrazine yielded benzaldehyde and vanillin pirazolines as white yellowish solid and white greenish solid in 45.89% and 34.15%, respectively. Antibacterial screening revealed that both vanillin chalcone and benzaldehyde pirazoline were not active as an antibacterial agent, while benzaldehyde chalcone was active for Gram Positive bacteria.Vanillin pirazoline was found to exhibits an antibacterial activity with the value inhibition zone/concentration againts Staphylococcus aureus (2.80/1000), Bacillus subtillis (2.25/100), Bacillus cereus (3.75/500), Escherichia coli (3.75/500) and Shigella flexneri (2.00/1000).
