- Background : Tuberculosis (TB) is a disease caoused by the bacterium Mycobacterium tuberculosis. Gold standard used in the diagnosis of tuberculosis is a bacterial culture. Gold standard bacterial culture performed in solid media Lowenstein-Jensen (LJ). The benefit of LJ solid medium is low cost but it took a long time. MGIT (Mycobacterium Growth Indicator Tube) is a growing medium for bacteria Mycobacterium sp. including M. Tuberculosis. the benefit of MGIT culture medium is fast time in diagnosing but susceptible to bacterial contamination. In this research the presence of bacterial contaminants in the medium MGIT resolved by administration of additional antibiotics in MGIT namely Gentamicin, Piperacilin, or a mixture. Objective : to determine the effect of antibiotic Piperacilin, Gentamicin, or mistures to the growth of Escherichia coli in cultures of Mycobacterium tuberculosis with MGIT liquid medium. Methods : subject of research were Mycobacterium tuberculosis and Escherichia coli were diluted from 108 Î1⁄4L/ml to 104 Î1⁄4L/ml. The samples used are divided into two groups, The Control Group (control M. Tuberculosis and control of Escherichia coli.) and Treatment Group (Piperacilin groups, Gentamicin groups, and mixed groups). Incubation at 37ÂoC, the turbidity observed visually and by using MGIT reader every day. Then planted in Mc.Concey and Gramâ��s staining to confirmed the presence of the growth of Escherichi coli. Ziehl Neelsen staining was also performed to confirmed the growth of Mycobacterium tuberculosis in MGIT tube. Observe the presence or not gram-negative bacterium bacillus (Mycobacterium tuberculosis) at 100x magnification under a microscope. Results : the results obtained the administration of Piperacilin can be used to reduce the growth of Escherichia coli in culture medium of MGIT. However, antibiotics Gentamicin and the misture is not be able to reduce the growth of Escherichia coli in MGIT medium.
