- Background: The liver is a vital organ in metabolism process that very susceptible to be damaged by free radical. As an antioxidant, xanthones in mangosteen peel extract (Garcinia mangostana L.) is expected to have a function as a hepatoprotective agent. Objective: To determine the effect of mangosteen peel extract (Garcinia mangostana L.) to the rate of hepatocyte damage after carbon tetrachloride (CCl4) induction. Methods: This study used a quasi-experimental design with posttest control group design. 30 Swiss mice (Mus musculus L.) were divided into 6 group. Group 1 (K1) as a normal control group got no intervention. Group 2 (K2) as a negative control group were induced by 20% carbon tetrachloride (CCl4) 1ml/BW/day. Group 3 as a positive control group were induced by 20% carbon tetrachloride (CCl4) 1ml/BW/day and got silymarin with 200mg/BW/day dose. Group 4 (K4), 5 (K5) and 6 (K6) worked as intervention group and induced by 20% carbon tetrachloride (CCl4) 1ml/BW/day and got mangosteen peel extract with dose variation as interventions. The doses are 150mg/BW/day for group 4 (K4), 300mg/BW/day for group 5 (K5) and 450mg/BW/day for group 6 (K6). The rate of hepatocytes damage measured by histopatologic examination with Haematoxylin � Eosin stain. The percentages of hepatocyte damage were taken by comparison between number of necrosed hepatocytes divided by total hepatocytes counted in 5 visual fields. ANOVA test was conducted to determine the effects of the interventions. Result: The rate of hepatocyte damage were decreased in all of intervention groups that statisticaly significant. The significance values are 0.002 (K4)
