  • Background: Malaria is one of the most prevalent tropical disease in the world. Until now, the use of antimalarials generated a lot of resistance that many scientists were developing research using herbal plant ingredients, for example mistletoe (D. pentandra). Previous studies have shown that D. pentandra have compound that has antimalarial activity. Objective: To determine in vitro antiplasmodial activity, mechanism of heme polymerization inhibition, and cytotoxicity of water extract of D. pentandra. Method: Antiplasmodium test conducted on P. falciparum using Giemsa staining. The heme polymerization inhibition test was conducted using hematin with test compounds using glacial acetic acid 100% and NaOH 0.1M. Cytotoxicity test performed on Vero cells that had been incubated with test compound. Result: The water extract of D. pentandra has antiplasmodial activity with IC50 values of 45,39Î1⁄4g/mL. The antiplasmodium mechanism of water extract occurs through inhibition of heme polymerization with IC50 values of 5.49Î1⁄4g/mL. The cytotoxicity IC50 values is 1394.4Î1⁄4g/mL. Selectivity index of water extract of D. pentandra leaves is 30,72. Conclusion: Dendrophthoe pentandra has in vitro antiplasmodial activity against P. falciparum through the mechanism of heme polymerization inhibition with low cytotoxicity.
