  • Nifedipine (NIF) is a cardiovascular drug which is frequently administrated several times a day. When administered orally in conventional solid dosage forms, NIF absorption is low. Therefore, it is necessary to develop sustained-release dosage form that can be retained in the stomach for a prolonged period known as Gastroretentive Dosage Form (GRDFs). With this system, NIF could be given once a day, as a consequence, the patient compliance and therapeutic target are assured. This research aimed determine the effect of the combination of HPMC K100M, xanthan gum and effervescent components on the tablet mass, physical properties of tablets, and the release of active substances. Besides, in this study, the composition of each factor producing the optimum formula was also evaluated. The research design used was 23 factorial design to obtain 8 formula. The optimization parameters used are floating lag time (FLT), absorption potency, C360 and DE360. Determination of the dissolution mechanism and dissolution profiles is based on the curve linearization method. Based on the desirability values, the optimum formula with HPMC K100M 81.0 mg, 48.2 mg of xanthan gum and 60,0 mg effervescent components were obtained. The evaluated parameters in optimum formula namely floating lag time, absorption potency and C360 are significantly not different to the those obtained from the software (Design Expert 7.1.3).
