- Flat nose viper snake (Trimeresurus puniceus) is one of the poisonous snake are often cause snake bite victims in humans. The venom of flat nosed viper snake (Trimeresurus puniceus) was a hemotoxin that destructing red blood cells or erythrocytes making the victim suffered severe animea into death. The venomâ��s concentration of this viper that caused fatality was unknown still. The aim of this research was to study the activity of the flat nosed viperâ��s venom on dogâ��s erythrocyte. The venom activity of flat nosed viper (Trimeresurus puniceus) tested in vitro. The venom collected in standard milking venom, keeps dried into crystal and weighed. The venom then diluted into PBS in concentration of 10%, 1%, 0,1%, 0,01%, 0,001%. The erythrocyte used in this study was in concentration of 20%. The venom activity was tested by pouring 10ÂμL of erythrocyte dilution with 10ÂμL of venom dilution for 20 seconds, then making a smear staining in Giemsa and observed microscopically for evaluating the damage of the erythrocytes. The result showed that venom dilution in 0,001% caused light abnormality on the erythrocyte dogâ��s especially on the wall of the some small erythrocytes becomes irregular. Venom dilution in 0,01%, 0,1% and 1% caused severe irregularity on the erythrocytes wall becomes crenation. Venom dilution in 10% caused more severe damages on the erythrocyte wall, making most of the populations of erythrocyte wall brakes and lysis. Based on the result, it could be concluded that flat nosed vipersâ��s venom having a high activity on destructing dogâ��s erythrocytes. Dogâ��s erythrocyte will get hemolysis when it is given venom with concentration â�¥10%. It need further research with more parameter of erythrocyte damages for more accurate results.
