  • Orally disintegrating tablets (ODT) is a development of pharmaceutical dosage form which is currently studied. ODT meloxicam (MEL) has problems about the low solubility and bitter taste of MEL. This study aims to determine the effect of complexation MEL-Ã�Â2-cyclodextrin to the solubility and taste masking of MEL which complexed by kneading method, the effect of Ac-Di-Sol and Kollidon CL on tablet disintegration time, and get superdisintegrants composition on optimum formula. Determination the formula is reached by the D-optimal mixture design program so that is obtained 5 formula by variation of Ac-Di-Sol and Kollidon CL concentration, distributed in 13 different runs. ODT MEL is made by the direct compression method. The properties of granul and tablet were compare with the literature of ODT dosage requirement and analyzed by the D-optimal program from Design Expert 7.1.5 software in order to obtain the optimal formula. The results show that complexation of MEL-Ã�Â2-cyclodextrin by kneading method can mask the bitter taste and increase the solubility of MEL from 0.0202 mg/minute to 0.0253 mg/minute in phosphate buffer pH 6,8. Kollidon CL can accelerate the disintegration time, while Ac-Di-Sol can prolong the disintegration time of ODT MEL. The formula consists of 4,5 mg Ac-Di-Sol and 10,5 mg Kollidon CL yields DE60 93,12%. The optimum formula consists of 5,4 mg Ac-Di-Sol and 9,6 mg Kollidon CL.
