- A Pseudomonas aeruginosa strain expressing azoreductase activity was utilized to degrade an azo dye Reactive Orange 86 (RO 86). The effect of various parameters such as agitation process, incubation time, the number of bacterial colonies, pH, and initial concentration of solution were investigated. The product resulting from bacterial degradation was analyzed with spectrophotometer UVvisible and compared with the initial spectra of dye solution. The results show that Pseudomonas aeruginosa can not decolorize RO 86 under agitation condition. The optimum degradation of RO 86 occurred after 96 hours incubation time and pH 6 under static incubation. The increase in the number of bacterial colonies enhanced the efficiency of degradation into optimum condition at 2,3 x 10 10 cfu of bacterial colonies, while the increase in the initial dye concentration lowered the efficiency of dye degradation. The UV-visible analysis of the compunds resulting from bacterial degradation suggests that the molecular structure of RO 86 changes after degradation and it can be inferred that the azo bond is cleaved during the degradation, but the dye is not completely degraded to be simpler molecules.
