- Improving orchids quality of Spathoglottis plicata Blume and Hybrids Vanda (Vandalimbata Blume X Vandatricolor Lindl.) has been carried out by polyploidy technique in vitro using colchicine. The aims of this study was to determine the effective concentration of colchicine in increasing ploidy levels of S. plicata Blumeand number of Protocorm Like-bodies (PLBs) of HybridsVanda. The variations of colchicine concentrations used for those two species of plants was different.For S.plicata Blume the concentrationwere 0.50%, 0.75%, 1.00%, 1.25%, 1.50%.For HybridsVanda the concentration were 0.01%, 0.03%, 0.05%, 0.08%, 0.1%, 0.5%,1.0% and 0,00% (as control).The duration of soaking for S. plicata Blumeduring 1, 2, 3, 24, 48 and 72 hours, while for Hybrids Vanda during 3, 6, 9 18 hoursand 2, 4, 7, 21 days.This studyused exsplant PLBs phase 2 leaves and was soaked in colchichine solutions + VW liquid medium with coconut water. Observations were performed every week, including: characters of morphology, physiology, anatomy and cytology. Chromosome preparation from root tips of S. plicataBlume was performed using squash method. Thecalculation chromosome number and the measurement of nucleus diameter were done under microscope connected to OptiLabProfessionalwith 1000x magnification. The results showed that the concentration of 0,5% colchicine for 24 hours effectively increased the chromosome number in PLBs of S. plicata Blume while concentration of 0,01% colchicine for 18 hours effectivelyincreased shoots number of PLBs ofHybrids Vanda twice as normal.In S.plicata Blume, thehigher concentration ofcolchicineinhibitedthe growth ofplant organs,increased thesize of stomata, the diameter ofcellnucleusandin contrastcanreducedthe index of stomata. The increasednumber ofS.plicata�s chromosomesreached 64 chromosomes.
