- Most nitrate reduction process in water is denitrification, using nitrate and nitrite which are subsequently converted into nitrogen gases. Denitrifying bacteria is a group of nitrate reductor, which has non-fermentative characteristics to degrade organic compounds under anaerob and aerob environmental condition. Utilization of nitrate as electron acceptor by this group bacteria may decrease nitrate level in water and produce N2O (nitrrous oxide) as a by product. Samples of mangrove sediment were obtained from Tritih mangrove habitat in Cilacap and Desa Semat, Jepara. Microbial isolates obtained from samples of mangrove sediment, were tested for their ability to reduce nitrate (NO3), and were analysed qualitatively (forming nitrate and gases) and quantitatively (ability to reduce nitrate). The purpose of this research was to obtain denitrifying bacteria and to find out their ability to reduce nitrate. The results of this study showed that the number of nitrate reducing bacteria from Cilacap was greater than the number from Jepara. Qualitative selection resulted in 38 isolates gave positive reaction to the Griess reagent, while the quantitative selection resulted in 11 demonstrated isolates of nitrate reduction. Statistical analysis showed that each isolate has a significant effect on the decreased levels of NO3.
