  • Gamavuton-0 (GVT) is an analog of curcumin with molecular formula 1,5-bis(4'- hidroksi-3'-metoksifenil)-1,4-pentadien-3-on. GVT-0 has better stability than curcumin under influence of pH and light. GVT-0 has biological activity as antiinflamatory, antioxidant, and antitumor agent. The weakness of GVT-0 is its physical property in water, where GVT-0 has low solubility in water so that the absorbsion of GVT-0 in G.I tract will be decreased, so do the bioavailability. One method that use in increasing the solubility of non polar molecule is by entrapping hydrophobic molecule in matrix of hydrophilic polymer such as chitosan and alginate in nanoparticle form. The purpose of this study is to attain the mechanism of forming GVT-0 nanopacticle in matrix of chitosan and alginate, and to observe the characteriztic and the stability of the nanoparticle. Preparation is done using the ionic gelation method with the variation of GVT-0 concentration in etanol 96% solution that mix with chitosan in acetic buffer and alginate solution in aquadest in ratio 1:1:1. The forming of nanoparticle occurs due to the ionic interaction among the components. Nanoparticle of GVT-0 is characterized by observation of its physical stability, the morfology under transmission electron microscope, % entrapment efficiency, observation of its stability in artificial gastric fluid (AGF) dan artificial intestinal fluid (AIF) in spectroscopy. This study has resulted 3 formulas that contain chitosan and alginate 0,01% in each formula, and variation in GVT-0 concentration 0,02%, 0,03%, and 0,04%. Observation its physical stability shows that all of the formula is free of precipitation, morfologyâ��s observation shows that the forming particle has size less than 100 nm and has spherical shaped. Results of measuring % EE of the formulas are 86,14±1,13%, 91,88±0,25%, 93,12±0,60. Observation of formulas stability in AGF shows great stability that all of the formulas can mantain more than 90% GVT-0 in each complex, in other hand, stability in AIF shows less than in AGF, where only the formula of GVT-0 0,04% can keep more than 80% formula in complex in 4 hours observation, the others formulas mantain less than 80%.
