  • Reproduction is one way to preserve the species living things. Gonads or testes is the male reproductive organs which is the site of spermatogenesis accured. Testicular activity is influenced by some factors, including environment, age and testosterone levels. Another hormone that is closely related to reproduction is thyroid causes hypothyroidism at low levels. Hypothyroid condition is one of the causes of sexual dysfunction. The purpose of this study is to determine the activity of the testicular of white rats (Rattus norvegicus Berkenhout, 1769) in hypothyroid induction. The research design were used Factorial Complete Randomized Design (FCRD) with two treatment factors based on hypothyroidism induction and age. The study used 48 male rats aged 4, 5, 6, and 7 weeks, divided into a control group and the group induced hypothyroidism using 0.05% propiltiourasil (PTU). Induced hypothyroidism conducted in two ways, ad libitum and oral gavage. Testicular activity analysis is performed by measuring testosterone levels using Enzyme Linked Immunosorbant Assay (ELISA) method, spermatogenesis activity examined by histological preparations using paraffin method, gonadosomatik indexes, and analysis of the quality of spermatozoa. The result of this study showed that induction of hypothyroidism with oral gavage was significantly reducing (p <0.05) levels of testosterone, spermatogenic cell number, percentage of viability, and increasing significantly (p <0.05) percentage of sperm morphological abnormalities. Induction of hypothyroidism by ad libitum over a significant effect (p <0.05) to decrease Gonadosomatik index and velocity of spermatozoa. Based on this study, it can be concluded that the hypothyroidism induction method by oral gavage giving an significant effect to decreasing gonad activity than ad libitum induction.
