PENGARUH APLIKASI MEMBRAN CARBONATED HYDROXYAPATITE (CHA) - GELATIN / KITOSAN TERHADAP PROLIFERASI FIBROBLAS (Kajian In Vitro)

- The body have a natural mechanism to repair the tissue damage caused of trauma or abnormalities. In the tissue regeneration, one of importance phase is fibroblast proliferation. It play a role in the process of synthesis, deposition and connective tissue remodelling. In previous study, the scaffold-based Carbonated Hydroxyapatite (CHA) was reported have a good biocompatibility, osteoconductivity and it could increased the local concentration of calcium and phosphate ions that was needed for new bone regeneration. The CHA mechanical properties should be improved related to material characteristic due its difficult clinical handling to take the powder particles to fill in bone defect without any disperse or migrate in other site. The solution of this case is to combine CHA powder with natural polymer such gelatin and chitosan. The aim of this study was to investigate the effect of the applications CHA-gelatin/chitosan membrane of fibroblast proliferation (in vitro). The subjects of this study were vero cell fibroblasts with concentration of 2x10 4 cells/ml. Vero cell fibroblasts were devided into 3 groups. The first group just for vero cell fibroblasts without membrane application (control group). The treatment groups were devided to CHA-gelatin membrane and CHAgelatin/ chitosan membrane application. Absorbance values or Optical Densuty (OD) of fibroblast proliferation was read using ELISA reader at 550nm after 12, 24, 48, and 72 hours time of observation (n=3). statistical analysis Two-way ANOVA showed that the application of CHAgelatin/ chitosan membrane and treatment duration influenced the fibroblast proliferation. Fibroblast proliferation in CHA-gelatin/chitosan membrane group was the highest than any other treatment groups in each subdivided time. At the 72 hours time observation, the result showed that there were significant increase fibroblast proliferation on CHA-gelatin/chitosan membrane group and CHAgelatin membrane group. In conclusion, the application of CHA-gelatin/chitosan membrane could increased the fibroblast proliferation.
