  • Fiber-reinforced composite (FRC) has recently been developed in dentistry. This material consists of polymer matrix and reinforcing fibers to increase the strength of the composite resin. E-glass fiber is one of the commonly used fiber in dentistry due to its good properties. The use of FRC in the oral cavity promotes a sustainable interaction between the FRC and saliva which is mostly consists of water. Polymer matrix absorbs water and facilitate a monomer release which can cause a cytotoxic effects to human cells. The purpose of this study was to determine the effect of E-glass FRC immersion duration in water against cytotoxicity on fibroblasts. The material used in this research are E-glass fibers (R338-2400/V/P. StickTech Ltd., Turku, Findland) and flowable composite resins (MasterFlow, Biodinamica, Brazil). Twenty five samples of E-glass FRC which sized 2 x 2 x 5mm immersed in water for 1, 7, 14, 21, and 28 days (5 samples each group). After the immersion was completed, the water extracts were added into the culture of fibroblast cells (Vero cell line). Cytotoxicity test were done by using the MTT method. The optical density was calculated using the ELISA plate reader to determine the percentage of surviving cells. The highest mean of cellâ��s viability resulted in day 1 (86.31% + 1.81) and the lowest mean of cellâ��s viability resulted in day 28 (84.11% + 2.81). The viability of fibroblasts after exposure to water were decreased from day 1 to day 28. The result of ANOVA test showed that the treatments given between groups was not significantly different (p> 0,05). The conclusion of this study: immersion duration of E-glass FRC up to 28 days has no significant effect against cytotoxicity on fibroblasts.
