  • Goat's milk has various peculiar properties that are good for health. One of the obstacle in dairy goat farming is the existence of an inflammatory disease of the udder called mastitis. This disease may cause highly economical loss for farmers. Most of the causes of mastitis is bacteria, and the main one is Staphylococcus. Staphylococcus are grouped into pathogenic (S. aureus, S. intermedius, S. hyicus) and non-pathogenic (S. epidermidis) bacteria. The identification to distinguish between pathogenic and non-pathogenic Staphylococcus depends on several phenotypic characters. This research was aimed to determine differences between pathogenic and non pathogenic Staphylococcus phenotype which was isolated from goat milk. A total of 28 isolates of Staphylococcus consist of 9 S. aureus, 9 S. intermedius, 4 S. hyicus and 6 S. epidermidis that have been isolated from ettawahybrid goatâ��s dairy milk were reidentified, and phenotypic characterization was applied including pigment/color of the colony and hemolytic behaviour on Blood Agar Plate (PAD) media, coagulation behaviour using plasma of rabbits, fermentation behaviour on Mannitol Salt Agar (MSA) and Vogel Johnson Agar (VJA) media along with acetoin production using Voges Proskauer test (VP). The results of this research showed that bacterial pathogenicity behavior grouping was based on MSA or VJA test. The character of pathogenic Staphylococcus was fermenting mannitol on MSA and it grew as black colonies surrounded by yellow zone on VJA media. The distinguishing test between S. aureus and S. intermedius depends on VP test which gave a positive result for S. aureus dan negative one for S. intermedius. Character of Non-pathogenic Staphylococcus was not fermenting mannitol on MSA and grew as black colonies on VJA. Based on the result of isolates characterization, it can be concluded that there was a mutation of the bacteria classification, including pathogenic and non-pathogenic Staphylococcus. The pathogenic Staphylococcus are S. aureus and S. intermedius whether the nonpathogenic ones are S. hyicus and S. epidermidis.
