- Anther culture can be used to obtain homozygous lines and accelerate the efficiency of selection. Japonica is subspecies with high anther culturability, while indica is recalcitrance. The research was intended to obtain information of medium composition and pre-treatment that suitable for anther culture in japonica and indica, and to develop the anther culturability using introgression anther culturability from japonica to the indica. The research was conducted from January 2009 until December 2012 in Plant Tissue Culture Laboratory, PT BISI International Tbk, Kediri, East Java. Nine genotypes were used as a source of anthers, represent japonica, indica, and F1 represent japonica � indica. The results showed that genotype affect double haploid induction through anther culture. Crossing between the two genotypes can give positive results in increasing response in anther culture of F1. Panicle pre-treatment in 4 oC for 8 days, using medium consist from modified N6 basal salt + NAA 2 mg/L + kinetin 0,5 mg/L + sucrose 54 g/L + putrescin 0,1644 g/L + Phytagel 2,5 g/L are suitable for japonica and hybrid derived from japonica � indica. Panicle pre-treatment in 4 oC for 9 days, using medium consist of modified N6 basal salt + 2,4-D 2.5 mg/L + kinetin 0,5 mg/L + AgNO3 10 mg/L + maltose 40 g/L + Phytagel 2,5 g/L and modified N6 basal salt + 2,4-D 2,5 mg/L + kinetin 0,5 mg/L + maltose 50 g/L + AgNO3 10 mg/L + Phytagel 2,5 g/L can enhance callus induction in �Ciuhao� and several genotype of indica. Five doubled haploid lines successfully obtained, crossing between japonica and indica can be effectively conducted to enhance response of hybrid to medium and increase the possibility to obtain doubled haploid line better than its indica parental line.
