  • The objective of the research was to identify the absence of bacteria isolates from a fermenting Indigofera tinctoria leafs which has the ability to reduce indigo compound to leuco indigo, to identify the genus of the indigo reducing bacteria isolates, and to obtain the reduction reaction parameters value of the indigo reducing bacteria. The indigo reducing bacteria from this research can be used as a natural alternative to Sodium Dithionite in the indigo dyeing process so that the environmental damage caused by the sulfur residue formed by Sodium Dithionite can be minimized. The indigo reducing bacteria isolated from a fermenting Indigofera tinctoria leafs using PYB medium supplemented with 0,01 % indigo, the ability to reduce indigo were examined. The indigo reducing bacteria purified by using a streak plate method using PYA-plate medium. Isolation of bacteria obtained 11 isolates which indicated indigo reducing bacteria. Genus characterization of the bacteria isolates done base on phenotypic characterization compared with others bacteria using a profile matching. From 11 isolates, five gram-positive isolates were choosen because the sustainable from the enviromental changes. Two isolates ST1 and ST8 were choosen among 5 gram-positive isolates. The identification shows that ST1 and ST8 belonged to genus Bacillus. The evaluation of the reduction reaction parameters value were carried out in a batch reactor at temperature 140C, 250C and 400C. Changes in the concentration of indigo (Cs) and bacteria (Cbio) in a batch reactor were determined periodically. Cs dan Cbio were analyzed by the OD, λ 620 nm used to analyzed Cbio, λ 380 nm used to analyzed leuco indigo concentration in the solution, by the standart curve fitting metods Cs were obtained. Values of Âμ0, kd, Ks, and Yi were evaluated by the curve fitting between values of Cs and Cbio resulting from mathematical simulation and values of Cs and Cbio observed in the experiments.Values of Âμ0, Ks, and Yi were evaluated at exponential phase. Values of kd were evaluated at stationary phase and death phase, using values of Âμ0, Ks, and Yi obtained from previous evaluation. The highest indigo compound reduction to leuco indigo by ST1 bacteria carried out at 250C, values of Âμ0 = 0,115 day-1, kd =0,07 day-1, Ks = 300 mg substrate/L, and Yi = 0,6 mg bacteria/mg substrate.
