- Halal issues related to contamination of pork is one thing that has always been a major concern and consideration in the food industry because of religion, health and economy reasons. Real-time PCR using primers specific species of ND5 mitochondrial DNA have conducted to identify the presence of porcine in processed meat like burger that mixed meat of pork and beef or chicken that combined with hot start method in fluoresence dye have been conducted to identify the presence of porcine in processed meat like burger. Various concentrations of pork added to the manufacture of beef and chicken burger from the range 0.5% -100%. The results showed that the contamination can be detected in all of pork samples that generate fragments of 227 bp amplification of pig mitochondrial ND5 gene. Detection limit of porcine was 1 pg and 0,5% in burger mixed with pork.
