- This study aims to get bacterial source of phytase, characterization of bacteria include gram stain, biochemistry and identification molecular, and characterization of phytase activity includes the optimization of temperature, pH, Km and Vmax. Bovine rumen fluid and feces as the source of isolates taken from a cow that fistulation. Phytase screening medium (PSM) used growth medium for isolation that containing Na-Phytat 0.5% as substrat and 1.8% agar that were grown anaerobically. Bacterial isolates obtained as many as six isolates derived from rumen isolation and 22 isolates from cattle feces isolation. Selection of phytase-producing bacteria using CoCl2 2% staining and ammonium molybdate and phytase activity assay. From the selection results were obtained 15 isolates of phytase-producing bacteria that can grow anaerob medium and than 15 isolates were grown in aerobic medium, obtained 14 isolates. Each of the isolates that grew in medium anaerobic and aerobic were measured of enzyme activity. The highest activity in bacteria grown in anaerobic and aerobic medium were chosen as the isolates selected for further test. F1 was selected isolate from feces. Phytase characterization of isolates F1 has the optimum temperature is 45oC, pH optimum 7.5, the value of Km is 14,94 uM and Vmax is 6,33 uM/second. Molecular identification 16S rRNA and biochemical of the isolate F1 homologous with Escherichia coli.
