- Banana (Musa spp.) is the most important fruit in almost every country, including Indonesia. Bananas can be infected by one or more viruses and the virus is able to infect multiple crops in one family or other family. The research was conducted to detect and molecular characterize of BBTV and BSV, and determine host range of BBTV. This research can be used for monitoring of epidemic and the virus evolution that is expected to become the basis for virus disease control strategy in bananas. Infected plants were collected from Lampung, Cianjur and Yogyakarta. Host range plant of BBTV from Cannaceae, Zingiberaceae, Heliconiaceae, and Araceae family. Phytopure DNA Extraction Kit method was used to extract the total DNA. Two pairs primers of BBTV (BBTV-F and BBTV-R) and BSV (BSV5466 BSV6196-F and-R) were used for Multiplex-PCR and DNA sequence analysis. BBTV and BSV PCR products were analyzed by RFLP technique using the restriction enzyme Dra I and Dde I, respectively. The results showed that BBTV and BSV DNA were successful amplified from samples of Lampung, Cianjur and Yogyakarta, and one sample from Yogyakarta was double infection. PCR detection of test host plant showed that Zinggiber sp. (Zingiberaceae) and Heliconia sp. (Heliconiaceae) as an alternative host of BBTV. The DNA sequence of BBTV samples from Lampung (LMP -20) and Cianjur (CJR-28) were identified as replicase gene of 1084 nt in length. The highest sequences homology that is the Bali isolates with amino acid sequence identity 99 % and 98.5 % respectively and included in the Asian Group. Sequence sample of YKT-A was identified as gene Reverse transcriptase and RNase H BSV is 709 nt in length. The highest sequence homology of sample that is China and SCBV isolate for amino acid sequence identity 94.9% and 86.4% respectively. RFLP analysis showed there was genetic diversity in some isolates of BBTV DNA 1 and genetic uniformity of RT and RH gene in BSV samples from Lampung, Cianjur, and Yogyakarta.
