- Metoprolol or 1 - [4 - (2-Methoxyethyl) phenoxy] -3 - [(1-methylethyl) amino]-2-propanol is a n antagonist of Ã�1-adrenergic receptor and is commonly used in the treatment of cardiovascular disorders such as hypertension, arrhythmia, and heart failure. Metoprolol has A 1% 1cm = 52a in aqueous acid at a wavelength of 274 nm which the molar absorptivity (? ) of 1390,48 L/(mol.cm). Metoprolol gave weak absorption in the UV (? ~ 1000 L/ (mol.cm)). This study aims to develop and validate a method to determine metoprolol concentration in spiked plasma by HPLC. Method used in this experiment based on the reaction between metoprolol and FDNB in acetonitrile. This solution was buffered with borate buffer at pH 10.0 and carried out at room temperature for 80 minutes. UV absorption of Metoprolol- DNB was measured at 249 nm. HPLC separation was performed by HPLC stationary phase Chromolith RP-18e (100 mm x 4.6 mm, 2 Âμm) and mobile phase of acetonitrile : acetate buffer 0.2 N pH 3.0 (30:70) with flow rate of 1. 0 mL/min. The method was validated based on the parameters of accuracy, precision, linearity, selectivity, LOD and LOQ. The linearity result at 300-1000 ng/mL obtained by standard curve equation y = 1.000 x +1051, with r = 0,9974. The LOD and LOQ are 79. 92 ng/mL and 266. 41 ng/mL respectively, while the% recovery from 95.27 to 102.29%. These results met the validation requirements.
