  • This study was investigate a group of soil fungi, especially yeasts associated with terrestrial orchid root system. P. susannae (L.) Raf. was used as objects of study. The orchid has a beautiful flower, has a specific habitat, and most numerous in the Forest Wonosadi. Specificity of orchids might be associated with less availability of specific nutrients in cooperation with soil microbes. Microbes, especially yeast found in many orchids rhizosfer. Several types of soil are cellulolytic yeasts that have significance in the procurement of certain nutrients for the orchid . The research objective was to obtain and analyze cellulose degradation activity by yeasts cellulolytic of soil yeasts rhizosfer orchid P. susannae (L.) Raf forms of cellulose. The study begin with the soil sampling rhizosfer the orchid root system of P. susannae (L). Raf. in two locations Wonosadi Forest. Soil samples were analyzed microbiology at the Laboratory. Isolation of cellulolytic yeasts performed using chytophaga media (CMC agar). Quantitative yeast isolates were selected based on its ability to grow on some form of cellulose (Cellulose Microcrystalline+, leaf litter and paper). Degradation activity by selected isolates performed by growing in liquid media chytophaga. Selected isolates were identified based on morphological characters and physiological and biochemical characters. The results showed that the four isolates obtained cellulolytic yeasts, namely K2, K16, A9 and A10. Degradation activity of the four isolates are the K2 (0.002 glucose/24 hour), K16 (0.011 glucose/24 hour), A9 (0.001 glucose/24 hour) and A10 (0.007 glucose/24 hour). Based on observations of morphological, physiological and biochemical character test conducted cellulolytic yeasts was allegedly a member of the genus Sacchromyces sp. (K2) and Candida sp. (K16, A9 and A10).
