- The previous studies about lactic acic bacterial (LAB) screening from gastrointestinal tract of broiler chicken were found 3 isolates probiotic candidate i.e: DB9 (duodenum), R01 and R02 (proventriculus). They were Gram positive bacteria, negative catalase, coccus, non motil, non gas producer, growth at 30, 37, 45 0 C, aerobic, anaerobic and agitation conditions. The viability test of LAB on acid pH, gastric juice (pepsin) and bile salt, antibacterial activity and biochemical identification (API 50 CHL kit) were studied, whereas molecular identification, organic acid profile, bacteriocin and its adherence on intestinal still unknown. The aim of this research were studied a taxonomical status of species level, organic acid profiles, molecular weight (MW) of bacteriocin and its adherence on mucus. Analysis of 16S rRNA sekuen was used as a based of molecular identification. HPLC was used for analysis of organic acid profiles. The ammonium sulphate precipitation and Tricine-SDS-PAGE was used for partial purification and separation of bacteriocin. LAB adherence on broiler mucus and porcine mucin was used microplate method with pepsin and bile salt stress conditions. Total plate count (TPC), absorbance and scanning electron micrograph (SEM) were observed as parameters. The results of this study revealed that R01, R02 and DB9 isolates based on sekuen 16S rRNA analysis have close related to Pediococcus acidilactici (99%) species. The three of LABs produced lactic acid (9.94 � 13.34 g/) and acetic acid (3.31 � 3.71 g/l). Bacteriocine was detected within 3.4 kDa of MW and was inhibited Salmonella pullorum ATCC 13036. All of P. acidilactici isolates adhere on broiler mucus with percentage of adherence and A550 were 55.5%
