  • As one of the richest country in biodiversity, Indonesia has a lot of animal and plant species. Tobacco (Nicotiana tabacum L.) is one of non-crop plants that drew up the diversity and were cultivated in almost all of areas in Indonesia. Madura Island, one of the centers for tobacco cultivation, has more than 70.000 ha or as much as 30- 33% of all tobacco cultivated area in Indonesia in 2006-2010. Wild species of tobacco have genes and phytochemistry that potential to be developed. However, information on the genetic diversity of Madura tobacco is still limited. This study was conducted to determine the genetic variation and relationship of 24 varieties of Madura tobacco with Random Amplified Polymorphic DNA (RAPD) and determine specific markers that define the characters of low-nicotine tobacco. Amplification of DNA with RAPD was started using 6 single primers (OPA- 18, OPB-12, OPB-14, OPC-1, OPC-8 and OPC-19) and 2 mixture primers ((OPB- 12+OPC-8) and (OPC-1+OPC-19)). Genetic similarity and clustering were analyzed by Unweighted Pair Group Method Arithmatic (UPGMA) method with Numerical Taxonomy and Multivariate Analysis System (NTSYS) version 2.10 software. Polymorphic loci resulting from amplification with double primers was relatively same with single primer (<60%) but with lower average of loci. RAPD generated specific markers of low-nicotine tobacco. Genetic variation of Madura tobacco was 0,2003 ± 0,1886 and the genetic similarity ranges from 77% â�� 100%. High similarity indicated close relationship among varieties of Madura tobacco. The dendogram showed that Madura tobacco varieties consist of 2 main clusters: cluster A (22 varieties) and cluster B (2 varieties: Bukabu Saâ��ang dan Prancak-95).
