- Indomethacin is a non-steroidal anti-inflammatory drug used in medicine under the trade name Confortid ® , Dialon ® and Reumatin ® . Indomethacin is heteroarilasetic acid analogue that has anti-inflammatory activity and analgesicantipyretic comparable to those of aspirin. Absorption of indomethacin can be improved by increasing the lipophilicity of this compound by converting the carboxyl group into the ester. The purpose of this study is to synthesize n-butyl indomethacin, 2-butyl indomethacin and isobutyl indomethacin and to determine the anti-inflammatory activity in vivo. The synthesis of compounds was carried out by esterification betwen indomethacin and n-butanol, 2-butanol or isobutanol respectively, using DCC as activator for 6 hours at room temperature. The product was extracted with ethyl acetate and purified by preparative thin layer chromatography. The Purity of the synthesized compounds were tested by TLC, melting point and gas chromatography, while this chemical structure were identified by, IR spectra, UV- Vis spectrometry , 1 H-and 13 C-NMR and mass spectrometry. The results have shown that the synthetic products are n-butyl indomethacin, 2-butyl indomethacin and isobutyl indomethacin with consecutive yield were 43,26%, 59,25% and 50,80%. The study anti-inflammatory used method by measured edema volume of rat feet from hour-0.5 until 4 hour with the help of Plethysmometer. Result showed that indomethacin, n-butyl indomethacin, 2-butyl indomethacin and isobutyl indomethacin at the same dose that is 10 mg/kg WB inhibited the inflammatory response: 33.59%, 46.92%, 72.15% and 52.29 % respectively.
