  • Diabetes mellitus is a disease caused by a disruption in the production of insulin by the pancreas that resulted in yperglycemia. One of inhibition mechanism of oral oglycemic drugs (OHD) is α-amylase inhibitor by blocking the digestion of starch in the intestine. An α-amylase inhibitors can slow the absorption of carbohydrates in the intestine. The purpose of the study was determine the α-amylase inhibitory activity of 96% ethanol extract of cashew leaf as an antidiabetic. Coarse powder cashew leaves was extracted by maceration method. An α-amylase inhibition analysis by the ethanol extract of cashew leaves carried by inhibition with soluble starch as a substrate that can inhibit the activity of α- amylase. The concentrations used 0,0625; 0,125; 0,25; 0,5 and 0.1 μM. Absorbance was read at λ 550 nm in the microplate reader. Kinetics analysis was done to determine the type of enzyme inhibition. Concentrations of kinetics analysis were consisted of 0.5; 1; 2; 4 and 8 μM. Absorbance read at λ 550 nm in the microplate reader. Result showed that the highest enzyme inhibition of α-amylase was obtained at 96.512% with an average IC50 of 17.011 μg/mL. Acarbose was positive control. The highest percent of inhibition was obtained at 76.263 % with an average IC50 of 9,640 μg/mL. The analysis enzyme inhibition kinetics of α-amylase by cashew nut leaf extract showed non-competitive mechanism.
