- Manilkara achras is one of tropical fruit plants used as tradisional drug. Based on the previous research ethanolic extract of Manilkara achras bark has antibacterial activity with concentration of 400μg/disk, 9 mm diameter of inhibition zone against S.aereus and 13 mm against E.coli. This study aimed to determine the antibacterial activity of ethanolic extract of Manilkara achras barks against multiresistant Escherichia coli and multiresistant Staphylococcus aureus and identify compound in the extract that has antibacterial activity. Ethanolic extract of sawo manila barks was tested for antibacterial activity using well diffusion methods with positive control kanamycin and negative control DMSO. Thin Layer Chromatography was done to determine class of compounds in the exstract a mobile phase of BAW:Ethyl acetate (2:8) and the stationary phase silica GF254nm followed by contact bioautography test. The test result showed that ethanolic extract of Manilkara achras bark has antibacterial activity, smallest concentration (625 μg/disk) provide inhibition zone 9,0 mm ± 0 against multiresistant E.coli and 8,33 mm ± 0,28 against multiresistant S.aureus. The highest concentration (5000 μg/disk) provide 12,66 mm ± 0,57 inhibition zone against multiresistant E.coli and 11,16 mm ± 0,28 against multiresistant S.aureus. The bioauthography test result shows that saponin compound responsible as the antibacterial against multiresistant E.coli and for the compound as antibacterial against multiresistant S.aureus is yet to be defined.
