  • One need to choose the right method to d etermine concentration of total dissolved protein in any particular condition or application. Here we report the analysis and co mparison between several methods used in determination of dissolved protein, i.e. 280 nm direct absorbance method, Biuret Assay, Lowry Assay, Bradford Assay and also BCA Assay. The effect of contaminants present in the sample, such as ammonium sulphate and sodium dodecilsulphate, to the protein determinat ion was also studied. Each particular method showed its own cha racteristics regarding sensitivity and susceptibility to the presence of certain contaminant. For general application, Bradford A%ay migh t obviously be the first choice of method, due to the procedure's simplicity. So far, Bradford Assay has been also proven to be a fast, sensitive, select ive and stable method in the presence of ammonium sulphate and SDS in the sample. The best linearity was also found in the Brdaford Assay.
