CINNAMON (Cinnamomum burmanii) ETHANOL EXTRACT HEPATOPROTECTIVE ACTIVITY AGAINST ISONIAZID INDUCED RATS LIVER HISTOPATHOLOGY

  • Salah satu obat yang dapat menyebabkan Drug induce liver injury (DILI) adalah obat antituberkulosis (OAT) seperti isoniazid (INH). Insidensi DILI akibat OAT di Indonesia adalah 22,5-43%. Kayu manis (Cinnamomum burmanii) memiliki manfaat sebagai antioksidan dan berpotensi sebagai hepatoprotektor. Penelitian ini bertujuan untuk mengetahui aktivitas hepatoprotektor ekstrak etanol kayu manis terhadap gambaran histopatologi hepar tikus jantan galur wistar yang diinduksi INH. Penelitian ini merupakan eksperimen laboratorium dengan posttest only control group design dengan jumlah sampel 25 ekor tikus. Hewan coba dibagi menjadi dari lima kelompok yaitu kelompok kontrol negatif yang diberi aquabidest, kontrol positif yang diinduksi INH dengan dosis 200mg/kgbb, dan tiga kelompok perlakuan yang dinduksi INH dengan dosis 200mg/kgbb dan diberi ekstrak etanol kayu manis dengan dosis 100, 200, dan 400mg/kgbb selama 14 hari. Pada akhir penelitian, hewan coba dikorbankan dan hepar diambil untuk kemudian dibuat preparat. Skoring histopatologi kerusakan sel hepar tikus menggunakan HAI-Knodell score. Hasil Knodell score dianalisis menggunakan uji One Way Anova dan Post Hoc Test Duncan. Hasil penelitian menunjukkan skor rata-rata tingkat kerusakan hepar tertinggi terdapat pada kelompok kontrol positif (2,80 ± 1,64), sedangkan skor rata-rata terendah pada kelompok kontrol negatif (0,40 ± 0,55). Skor kerusakan hepar kelompok yang diberi ekstrak etanol kayu manis dosis 100, 200, dan 400mg/kgbb adalah 1,20 ± 0,84; 0,80 ± 0,84 dan 0,80 ± 0,84 secara berurutan. Terdapat perbedaan yang signifikan antara kelompok kontrol positif dengan kelompok perlakuan (p<0,05), tetapi tidak terdapat perbedaan yang signifikan antar kelompok perlakuan.   • One of the drugs that can cause Drug Induced Liver Injury (DILI) is an antituberculosis drug such as isoniazid (INH). The incidence of DILI due to OAT in Indonesia is 22.5-43%. Cinnamon (Cinnamomum burmanii) has benefits as an antioxidant and potential as a hepatoprotector. This study aims to determine the hepatoprotector activity of cinnamon ethanol extract on the histopathological appearance of the liver of INH-induced Wistar male rats. This study was a laboratory experiment with a posttest only control group design with a sample of 25 rats consisting of five groups: the negative control group that was given aquabidest, the positive control group which was induced by INH at a dose of 200 mg/kgbw, and the three treatment groups which were induced by INH with dose of 200 mg/kgbw and given cinnamon ethanol extract at doses of 100, 200, and 400 mg/kgbw for 14 days. At the end of the study, the experimental animals were sacrificed and their livers were taken for preparations. Histopathological scoring of rat liver cell damage using the HAI-Knodell score. The results of the Knodell scores were analyzed using the One Way Anova test and Duncan's Post Hoc Test. The results showed that the highest average score of liver damage was found in the positive control group (2.80 ± 1.64), while the lowest average score was in the negative control group (0.40 ± 0.55). The score of liver damage in the group given cinnamon ethanol extract doses of 100, 200 and 400 mg/kg body weight was 1.20 ± 0.84; 0.80 ± 0.84 and 0.80 ± 0.84 respectively. There was a significant difference between the positive control group and the treatment group (p<0.05), but there was no significant difference between the treatment groups. Cinnamon ethanol extract has hepatoprotector activity with an effective dose of 100 mg/kgbw.
