- Abstrak. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi amonium sulfat terhadapakivitas protease ekstraseluler Lactobacillus plantarum B1765 isolat bekasam, serta menentukan aktivitas spesifik enzim hasil pengendapan mengunakan amonium sulfat dan dialisis. Penelitian ini dilakukan melalui dua tahap penelitian yaitu penentuan waktu akhir fase log pada kurva pertumbuhan Lactobacillus plantarum B1765 menggunakan metode turbidimetri dan pengendapan protease ekstraseluler Lactobacillus plantarum B1765 menggunakan berbagai konsentrasi amonium sulfat yaitu 15%, 30%, 45% dan 60%. Hasil penelitian menunjukkan bahwa waktu akhir fase log Lactobacillus plantarum B1765 adalah jam ke 16. Pengaruh konsentrasi amonium sulfat terhadap aktivitas protease adalah meningkatnya protease pada pengendapan menggunakan konsentrasi amonium sulfat 15% sampai 45%.Selanjutnya menurun pada konsentrasi amonium sulfat 60%. Aktivitas tertinggi diperoleh pada pengendapan menggunakan konsentrasi amonium sulfat 45% yaitu 0,697 Unit/mL dengan kadar protein sebesar 0,880 mg/mL sehingga diperoleh aktivitas spesifik sebesar 0,794 Unit/mg. Setelah proses dialisis, aktivitas proteasemengalami peningkatan menjadi 0,877 Unit/mL dengan kadar protein sebesar 0,729 mg/mLsehingga diperoleh aktivitas spesifik sebesar 1,202Unit/mg. This study aimed to know the effect of ammonium sulphate concentration toward protease extracelluler activity Lactobacillus plantarum B1765 of bekasam isolate, and determine specific activity of enzyme precipitation result use ammonium sulphate and dialysis. This study consists of two steps. They were determination the final time of log phase in growth curve Lactobacillus plantarum B1765 use turbidimetry methode and protease perecipitation use ammonium sulphate in various concentration which concentration were 15%, 30%, 45% and 60%. The result showedthat the time ofthe end log phase ofLactobacillusplantarumB1765is at the16 hour. The effect ofammoniumsulphateconcentration is the increasing ofproteaseactivity at precipitate use 15% until 45% ammonium sulphate concentration. Then decrease at 60% ammonium sulphate concentration.The highest activity is obtained at precipitate use 45% ammonium sulphate concentration is 0,697 Unit/mL with protein concentration is 0,880 mg/mL then specific activity is 0,794 Unit/mg. After dialysis, protease activity increase become 0,877 Unit/mL with protein concentration is0,729 mg/mLthen specific activity is 1,202Unit/mg.
