- ABSTRACTMiana leaf (Coleus scutellarioides [L] Benth.) contained flavonoids, tannins, triterpenoids,steroids and essential oils are able to provide an antibacterial effect. The purpose of the studywas to made a balm extract from Miana leaf (Coleus scutellarioides [L] Benth.) and test theeffectivenen against scar infected bacteria in rabbit skin. Proces of balm extract from Mianaleaf has qualified the standard and tested on rabbit with 5 treatment groups, is injuries withoutnegative control, positive control, Miana leaf ointment 20%, 40% and 80%. All the rabbit cutfor 1,5 cm and given 0,2 mL of bacteria Staphylococcus aureus. The wounds were appliedwith ointment three times daily. Observation was conducted everyday for 8 days. All datawas tested statistically using ANOVA (Analysis Of Variant) followed bay LSD (LeastSignificant Difference) test. The result shows differences that the Miana leaf formulationmeets the ointment test requirement according to Farmakope Indonesia Edition III, woundswere narrowed, scabs were formed and then wounds were closed. Statistical test showed thatthere were significant effects on wound healing in rabbit, F count > F table (37,167 > 5,19).Key words : ointment, Coleus scutellarioides [L] Benth, Staphylococcus aureus, rabbitABSTRAKDaun Miana (Coleus scutellarioides [L] Benth) memiliki kandungan flavonoid, tanin,triterpenoid, steroid dan minyak atsiri yang mampu memberikan efek antibakteri. Penelitianini bertujuan untuk membuat salep dari ekstrak daun Miana (Coleus scutellarioides [L]Benth.) dan uji efektivitas sediaan salep terhadap pengobatan luka yang terinfeksiStaphylococcus aureus pada kulit kelinci. Pembuatan salep ekstrak daun Miana yang telahmemenuhi syarat dan diujikan pada hewan uji kelinci sebanyak 5 ekor dengan perlakuan,dasar salep berlemak (kontrol negatif), Gentamicin salep (kontrol positif), salep daun Miana20%, 40% dan 80%. Semua kelinci disayat sepanjang 1,5 cm dan diberikan bakteriStaphylococcus aureus sebanyak 0,2 mL. Luka diolesi tiga kali sehari dengan salep yangdiuji. Pengamatan luka dilakukan setiap hari selama 8 hari. Semua data diuji secara statistikmenggunakan ANOVA (Analysis Of Variant) dan dilanjutkan dengan uji LSD (LeastSignificant Different). Hasil penelitian menunjukkan formulasi salep ekstrak daun Mianamemenuhi persyaratan uji salep menurut Farmakope Indonesia edisi III. Luka sayat yangdioleskan salep ekstrak daun Miana mengalami penyempitan luka, membentuk keropeng danmenutup luka mulai hari ke-3 (tiga). Hasil uji statistik menunjukkan efek signifikan terhadappenyembuhan luka pada kelinci, yaitu F hitung > F tabel (37,167 > 5,19).Kata kunci : Salep, Coleus scutellarioides [L] Benth, Staphylococcus aureus, kelinci
