EFEK PEMBERIAN GEL GETAH BATANG TANAMAN PISANG SECARA TOPIKAL TERHADAP KEPADATAN SERABUT KOLAGEN PADA PROSES PENYEMBUHAN LUKA PASCA EKSTRAKSI GIGI MARMUT

  • Abstract Banana stem sap contain of saponin, flavonoid, vitamin C, and tannin play a role in the wound healing process. The aim of this study was to determine the effect of topical application of banana stem sap gel on the density of collagen fiber in the wound healing process after tooth extraction in the guinea pig (Cavia cobaya). Fifty four guinea pigs were divided into three groups, each group consisted of 18 guinea pigs. Tooth extraction was performed on the mandibular left central incisive. The positive control group was treated with iod glycerin, and the negative control group was treated with CMC-Na. Subjects in the treated group were treated with 80% banana stem sap gel. After the treatment, three of each group were sacrified in 1, 3, 5, 7, 14, and 24 days after tooth extraction and the mandibles were collected. The mandibles were prossed for histological treatment prior to Mallory staining. Collagen fiber was scored under light microscope examination and then statistically analyzed by using Kruskall-Wallis and Mann-Whitney Tests. The result showed significant difference of collagen fiber density between treated and control groups (p < 0.05). The increasing of collagen fiber showed significant difference in 3, 5, 7, and 14 days after tooth extraction between treated groups with negative and positive control. In conclusion, topical application of 80% banana stem sap gel could promote deposition of collagen fiber in socket healing process after tooth extraction in guinea pig. Key words: banana stem sap gel, tooth extraction, collagen fiber Abstrak Getah batang pisang mengandung saponin, flavonoid, vitamin C, dan tanin yang berperan dalam proses penyembuhan luka. Penelitian ini bertujuan untuk mengetahui efek gel getah aplikasi topikal gel getah batang pisang raja terhadap kepadatan serabut kolagen pada proses penyembuhan luka pasca pencabutan pada soket gigi marmut (Cavia cobaya). Lima puluh empat marmut dibagi menjadi 3 kelompok, masing-masing terdiri atas 18 marmut. Pencabutan gigi marmut dilakukan pada gigi insisivus sentralis kiri rahang bawah. Kelompok kontrol positif diperlakukan dengan iod gliserin dan kelompok kontrol negatif dengan CMC-Na. Subjek pada kelompok perlakuan diberi gel getah batang pisang 80% secara topikal. Setelah perlakuan, pada 3 marmut dari masing-masing kelompok dilakukan dekapitasi pada 1, 3, 5, 7, 14, dan 24 hari pasca pencabutan gigi. Rahang bawah marmut dipotong lalu dibuat preparat histologis untuk pengecatan Mallory. Kepadatan serabut kolagen diamati dengan mikroskop cahaya, kemudian data dianalisis dengan Uji Kruskall-Wallis dan Mann-Whitney. Hasil penelitian menunjukkan ada perbedaan bermakna serabut kolagen (p < 0,05) antara kelompok perlakuan dan kontrol. Peningkatan serabut kolagen menunjukkan perbedaan yang signifikan pada 3, 5, 7, dan 14 hari pasca pencabutan gigi antara kelompok perlakuan dengan kontrol negatif dan positif. Kesimpulannya, aplikasi topikal gel getah pisang 80% dapat mempercepat pembentukan serabut kolagen pada proses penyembuhan luka soket gigi marmut pasca pencabutan gigi marmut.
