Regenerasi Sel Pulau Langerhans Pada tikus Putih (Rattus Norvegicus)diabetes yang diberi Rebusan Daging mahkota (Phaleria macrocarp Lam)

  • Background: Decocted pulp of Phaleria macrocarpa contains the active compound which have hypoglycaemic effect similar with insulin, and flavonoid by changing the Ca2+ metabolism and regenerating cell ? of islet of langerhans pancreas. The objective of this research is to investigate the ability of decocted pulp of Phaleria macrocarpa to decrease the level of blood glucose, and to regenerate the Langerhans islet of diabetic white rats. Design and Method: Twenty-five male white rats (Rattus norvegicus), weigh 120-180 gram, were inducted by alloxan as diabetogenic agent then divided to 5 treatment group, that is are negative control, positive control, groups treated by decocted pulp of Phaleria macrocarpa 4,5%, 9%, and 13,5%. The level of blood glucose was checked every day from 1st to 2nd week, then continued to 4th week. After treatment, all rats in each group were decapitated then the pancreas was taking to be analyzed by counting the number of Langerhans islet cells. Result: There was a significant difference in the number average of Langerhans islet cells between control group, groups treated by decocted pulp of Phaleria macrocarpa 4,5%, 9%, and 13,5% (Fcount=31.883>Ftable=2.50). Decocted pulp of Phaleria macrocarpa contributes to 43.82 % decreasing of blood glucose (r=43.82) and there was a strong relation between decocted pulp of Phaleria macrocarpa and the decreasing of blood glucose level average (r=0.66). Moreover, There was a significant difference in number average of Langerhans islet cells between control group, groups treated by decocted pulp of Phaleria macrocarpa 4,5%, 9%, and 13,5% (Fcount=22.441>Ftable=2.45). The number average of Langerhans islet cell was increase started at concentration of decocted pulp of Phaleria macrocarpa 9%. Conclusion: Decocted pulp of Phaleria macrocarpa can regenerate cell ? of islet of langerhans pancreas.?Pendahuluan: Daging buah mahkota dewa mengandung senyawa kimia aktif yang diduga mempunyai efek hipoglikemik mirip insulin yaitu flavonoid dengan cara merubah metabolisme Ca2+ dan meregenerasi pulau Langerhans pankreas terutama sel. Tujuan penelitian ini adalah untuk mengetahui pemberian rebusan buah mahkotadewa terhadap penurunan kadar glukosa darah, besar dosis pemberian rebusan daging buah mahkotadewa dan mengetahui adanya regenerasi sel pulau Langerhans pankreas tikus diabetes yang telah diberi rebusan daging buah mahkota dewa. Metode Penelitian: Duapuluh lima tikus putih jantan (Rattus norvegicus) dengan berat 120-180 gram yang telah diinduksi aloksan sebagai agen diabetogenik dibagi 5 kelompok perlakuan, yaitu kontrol positif dan negatif dan pemberian rebusan daging buah mahkotadewa 4,5%, 9%, dan 13,5%. Glukosa darah diperiksa pada saat dimulai penelitian, minggu ke-2 dan ke-4. Akhir penelitian hewan coba didekapitasi,diambil pankreas untuk diamati jumlah sel pulau Langerhans yang selanjutnya diuji statistik. Hasil Penelitian: Hasil penelitian? menunjukkan perbedaan bermakna antara kadar rata-rata glukosa pada perlakuan kontrol, pemberian rebusan daging buah mahkota dewa 4,5%, 9% dan 13,5 % (Fhitung = 31,883 > Ftabel= 2,50). Penurunan kadar glukosa darah mulai terjadi pada pemberian rebusan buah mahkotadewa 4,5%. Rebusan buah mahkota dewa memberikan kontribusi 43,82% dalam penurunan glukosa darah (r? = 43,82%) dan terdapat hubungan yang kuat antara mahkota dewa dengan penurunan kadar glukosa darah (r = 0,66). Terdapat perbedaan bermakna antara jumlah rata-rata sel pulau Langerhans pada perlakuan kontrol, pemberian rebusan daging buah mahkota dewa 4,5%, 9% dan 13,5% ( Fhitung = 22,441 > Ftabel = 2,45). Jumlah rata-rata sel Pulau Langerhans terdapat peningkatan mulai pada pemberian rebusan buah mahkota dewa 9%. Kesimpulan: Daging buah mahkota dewa dapat meregenerasi sel pulau Langerhans tikus putih diabetes
