- Background: Sarang semut (Myrmecodia pendens) is a traditional plant that has been known to containanti cancer components. Flavonoids and tannins were contained in sarang semut plant which are believedhas cytotoxic effect against cancer cell line. This study aims at cytotoxic effect ethanolic extract of sarangsemut at various concentrations on HeLa cervical cancer cell line.Design and Method: The method was quasi experimental with post test only non equivalent control groupdesign. HeLa cell was divided into two groups. The first group as positive control with doxorubicin, secondgroup as treatment with ethanolic extract of sarang semut at various concentrations. Ethanolic extract ofsarang semut concentrations used were 3,91 ?g/ml; 7,81 ?g/ml; 15,63 ?g/ml; 31,25 ?g/ml; 62,50 ?g/ml;125 ?g/ml; 250 ?g/ml; 500 ?g/ml; 1000 ?g/ml. Cytotoxic effect was evaluated by direct counting methodwith tryphan blue dye then using probit regression analysis to find IC50 value.Result: Inhibitory concentration 50 (IC50) value ethanol extract of sarang semut was 33,28 ?g/ml. Ethanolextract of sarang semut had a cytotoxicity effect categorized as the moderately active (20 ?g/ml< IC50< 100?g/ml). Inhibitory concentration 50 (IC50) value doxorubicin was 5,56 ?g/ml. Cytotoxicity effect of doxorubisinhigher than cytotoxicity effect of ethanolic extract of sarang semut.Conclusion: Ethanolic extract of sarang semut (Myrmecodia pendens) had a cytotoxic effect categorized asthe moderately active on HeLa cell (Sains Medika, 3(2):112-120).Key words: HeLa cervix cancer cell line, cytotoxic effect, ethanolic extract of sarang semut(Myrmecodia pendens).Pendahuluan: Sarang semut merupakan tanaman tradisional yang diketahui mengandung komponenanti kanker. Flavonoid dan tanin yang terkandung dalam tumbuhan sarang semut dipercaya memilikiefek sitotoksik terhadap sel kanker. Penelitian ini bertujuan untuk mempelajari efek sitotoksik ekstraketanol sarang semut (Myrmecodia pendens) dengan berbagai konsentrasi terhadap sel HeLa (sel kankerserviks yang dikulturkan) secara in vitro.Metode Penelitian: Penelitian kuasi eksperimental dengan post test only non equivalent control groupdesign ini menggunakan sel HeLa yang dibagi menjadi dua kelompok, yaitu kontrol positif dengandoksorubisin dan kelompok perlakuan dengan ekstrak etanol sarang semut dalam berbagai konsentrasi.Konsentrasi ekstrak etanol sarang semut yang dipakai yaitu 3,91 ?g/ml; 7,81 ?g/ml; 15,63 ?g/ml; 31,25?g/ml; 62,50 ?g/ml; 125 ?g/ml; 250 ?g/ml; 500 ?g/ml; 1000 ?g/ml. Efek sitotoksik dinilai dengan metodedirect counting dengan pewarnaan trypan blue kemudian dilakukan analisis regresi probit untuk mencarinilai IC50.Hasil Penelitian: Nilai IC50 ekstrak etanol sarang semut sebesar 33,2819 ?g/ml. Ekstrak etanol sarangsemut memiliki efek sitotoksik kategori cukup aktif (20 ?g/ml<IC50<100 ?g/ml). Nilai IC50 doksorubisinsebesar 5,5595 ?g/ml. Efek sitotoksik doksorubisin lebih besar daripada efek sitotoksik ekstrak etanolsarang semut.Kesimpulan: Ekstrak etanol sarang semut (Myrmecodia pendens) memiliki efek sitotoksik kategori cukupaktif terhadap sel HeLa (Sains Medika, 3(2):112-120).Kata kunci : sel kanker serviks HeLa, efek sitotoksik, ekstrak etanol sarang semut (Myrmecodia pendens).
