ISOLASI, IDENTIFIKASI, DAN UJI SITOTOKSIK SENYAWA ALKALOID DARI DAUN MINDI (Melia azedarach L.)

- Mindi (Melia azedarach L.) from Meliaceae tribe is one kind of plant that has several classes of compounds, one is alkaloid. The Mindi leaf has been extracted using a solvent of 96% ethanol by maseration method for elucidation of its fitochemistry. Alkaloid can be isolated from methanol-water extract of mindi leaf by salting extracted with ethyl acetate eluent. Separation of alkaloids was performed by preparative TLC using the eluent ethyl acetate: chloroform (6:4) and test its purity by TLC method using eluent single variety, mix, and two-dimensional. Structure analysis of the alkaloid isolates were carried out using UV-Vis spectrophotometer, FT-IR, and LC-MS as well as test cytotoxic alkaloid extrac usingt BSLT (Brine Shrimp Lethality Test) method. Identification of the alkaloid isolates using UV-Vis spectrometry showed that indole-group alkaloid has wavelengths of 220 nm and 270 nm. Analysis using FTIR spectrometry showed that alkaloids isolate has a functional group N-H, O-H, CH3, CH2, C = C aromatic, C-N and C = O. The isolat has a molecular weight of 290.45 g / mol by analysis using LC-MS. The cytotoxic test showed that LC50 of alkaloid extract of 21.273 ppm, so that it can be concluded that the alkaloid extract is highly cytotoxic.
