- ABSTRACT Dewandaru (Eugenia uniflora L.) has been reported having several activities such as antioxidant, antimutagenic,antitumor and inhibit DNA polymerase on EVB (Eipstein-Bar Virus). This research aimed to know the cytotoxiceffect of dewandaru leaves extract on breast cancer cell line, T47D and MCF-7. The 70% ethanolic extract ofDewandaru was tested to the breast cancer cell line of T47D and MCF-7 by MTT procedure on the concentrationsare as follows: 31,25; 62,5; 125; 250; 500 μg/ml. The chemical content of the extract was determined byphytochemical screening method. The result showed that cytotoxic activities (IC50 value) of dewandaru extracton breast cancer cell line of T47D was 117μg/mL while on MCF-7 was 155 μg/ml. The phytochemical screeningshowed that dewandaru extract contain chemical compound such as alkaloida, saponin, flavonoide, tanin andpolyfenol.Key words: Eugenia uniflora L., breast cancer, T47D and MCF-7. ABSTRAK Tanaman dewandaru (Eugenia uniflora L.) telah dilaporkan memiliki aktivitas antioksidan, antimutagenik,antitumor dan mampu menghambat DNA polymerase pada EBV(Eipstein-Bar Virus). Penelitian ini bertujuanuntuk mengetahui efek sitotoksik daun dewandaru (Eugenia uniflora L.) terhadap sel kanker payudara T47Ddan MCF-7. Serbuk daun dewandaru dimaserasi dengan penyari etanol 70%. Ekstrak kemudian diujikanterhadap dua macam sel kanker mengunakan metode MTT dengan konsentrasi 31,25;62,5 ;125 ;250 ;500μg/ml. Untuk mengetahui kandungan golongan kimia, dilakukan skrining fitokimia terhadap ekstrak daundewandaru. Dari uji sitotoksik ekstrak daun dewandaru terhadap sel kanker payudara T47D diperoleh nilaiIC50 sebesar 117 μg/ml sedangkan terhadap sel kanker payuadara MCF-7 diperoleh nilai IC50 sebesar 155 μg/ml. Dari hasil skrining fitokimia diketahui bahwa ekstrak daun dewandaru mengandung alkaloid, saponin,flavonoid, tanin dan polifenol.
