  • Abstract. Anopheline species confirmed as malaria vector if the salivary gland contained sporozoites. One of the method to confirmed it was through an Enzyme-Linked Immunosorbent Assay (ELISA). The aim of this study was to investigate the presence of circum sporozoite protein (CSP) in the mosquito of Anopheles barbirostris with ELISA method. The study was conducted in malaria endemic area named Modu Waimaringu Village, Waikabubak District, Sumba Barat Regency in March 2011. The study design was cross-sectional study, mosquito for the ELISA test were collected only from animal bait. ELISA method examination used on An. barbirostris body parts (i.e. the head-thorax) where sporozoites of P. falciparum or P. Vivax possibly be found. The results showed that 40 samples of An. barbirostris mosquitoes which acquired from the mosquite bait in Modu Waimaringu Village was negative (100%). It means that there was no CSP found and An. barbirostris was not a malaria vector in the area. Key words: circum sporozoite protein, Anopheles barbirostris, ELISA Abstrak. Nyamuk Anopheles spp. dinyatakan sebagai vektor malaria apabila ditemukan sporozoit di kelenjar ludahnya dan salah satu metode yang dapat dilakukan adalah melalui uji Enzyme-Linked Immunosorbent Assay (ELISA). Penelitian ini dilakukan untuk mendeteksi protein circum sporozoite pada tersangka vektor malaria Anopheles barbirostris melalui metode ELISA. Penelitian ini dilakukan di daerah endemis malaria di Desa Modu Waimaringu, Kecamatan Waikabubak, Kabupaten Sumba Barat pada bulan Maret 2011. Desain studi penelitian ini adalah cross sectional, nyamuk uji diperoleh melalui penangkapan nyamuk sekitar kandang. Uji ELISA dilakukan pada bagian kepala dan dada nyamuk An. barbirostris yang potensial mengandung sporozoit Plasmodium falciparum dan Plasmodium vivax. Hasil penelitian menunjukkan dari 40 sampel An. barbirostris yang diuji di Desa Modu Waimaringu seluruhnya negatif (100%). Hal ini berarti tidak ditemukannya protein circum sporozoite dan An.barbirostris bukan vektor pada daerah tersebut. Kata Kunci: protein circum sporozoite, Anopheles barbirostris, ELISA
