  • [<b>ABSTRAK</b><br> Latar belakang : Penyakit periodontal baik periodontitis kronis maupun periodontitis agresif adalah manifestasi klinis dari interaksi agen, yaitu bakteri seperti A.actinomycetemcomitans ( Aa ) dan produknya antara lain Cytolethal Distending Toxin ( CDT ) dengan sel inang di jaringan periodontal seperti sel fibroblas dan osteoblas. Interaksi tersebut dan dampaknya dalam patogenesis penyakit periodontal belum sepenuhnya dipahami. Tujuan penelitian : Untuk mengungkapkan peran CDT-like protein Aa yang diisolasi dari periodontitis kronis dan agresif dalam patogenesis penyakit periodontal, dengan cara memaparkan lisat bakteri yang mengandung CDT-like protein dengan fibroblas dan osteoblas. Dampak dari interaksi tersebut dievaluasi berdasarkan analisis viabilitas dan ekspresi mRNA IL ? 1, IL ? 6, TNF ? &#945; dan RANKL dengan menggunakan metode MTT assay dan Real Time PCR. Metoda : Digunakan tiga macam konsentrasi CDT-like protein dari lisat bakteri Aa hasil isolasi dari periodontitis kronis dan agresif yaitu 2 &#61549;g / &#61549;l, 10 &#61549;g / &#61549;l dan 20 &#61549;g / &#61549;l untuk uji viabilitas fibroblas dan osteoblas hasil kultur. Untuk uji ekspresi mRNA, digunakan konsentrasi 2 &#61549;g / &#61549;l CDT-like protein dari lisat bakteri yang sama dan diaplikasikan ke sel fibroblas dan osteoblas Hasil : Terdapat kecenderungan berbeda pada viabilitas dan ekspresi sitokin antara sel fibroblas dan osteoblas setelah diberi pemaparan dengan CDT-like protein Aa, baik yang diisolasi dari periodontitis kronis maupun dari peridontitits agresif.Kesimpulan : CDT-like protein yang diproduksi oleh dua isolat Aa yang berbeda yaitu dari periodontitis kronis dan periodontitis agresif mengakibatkan respons biologis yang berbeda terhadap sel fibroblas dan osteoblas.
